چکیده

Wax, Freese & Cashel (1967), Vary & Kornberg (1970), Dring & Gould (1971) and Foerster (I 972) have isolated mutants of Bacillus which germinate more slowly than the wild type. We have isolated slow germination mutants from an anaerobe, Clostridium bifermentans, and have shown that treatment with sodium hydroxide will increase their germination rates over Iooo-fold to rates similar to those of the parent (Wyatt & Waites, 1971). Dithiothreitol-urea, like sodium hydroxide, removes protein from spore coats (Aronson & FitzJames, 1971 ; Waites, Wyatt & Arthur, 1972). We report the effect of sodium hydroxide or dithiothreitolkea on these mutants.

Spores of Clostridium bifermentans germinated in the presence of L-alanine and sodium ions. The rate of germination was not altered by the addition of D-alanine but was increased by L-a-amino-n-butyrate, L-lactate, Lor D-arginine, L-histidine, DL-ornithine, L-lysine, pyruvate, Lor D-serine, L-phenylalanine or L-proline and decreased by D-phenylalanine, L-isoleucine, glycine or L-threonine. Germination of Bacillus megaterium spores after various extraction procedures.

The initiation of germination of Bacillus megaterium QM B1551 spores, grown in supplemented nutrient broth, has been studied. The initiation properties depend on buffer concentrations and the particular batch of spores. Initiation in l-alanine, KBr, calcium dipicolinate, or in buffer alone increases as a function of the spore age; whereas initiation in glucose, l-leucine, or l-proline remains r...

Production of Clostridium bifermentans Spores as Inoculum for Bioremediation of Nitroaromatic Contaminants.

Spores of Clostridium bifermentans KMR-1 were produced for use as a microbial inoculum for bioremediation and were preserved in both liquid and dry forms. All spore formulations showed good viability and ability to biodegrade the target compound, 2,4,6-trinitrotoluene (TNT), after 4 months of storage. For low-cost bulk spore production, several medium compositions, based on soy peptone, corn st...

Growth from spores of Clostridium perfringens in the presence of sodium nitrite.

The method by which sodium nitrite may act to prevent germination or outgrowth, or both, of heat-injured spores in canned cured meats was investigated by using Clostridium perfringens spores. Four possible mechanisms were tested: (i) prevention of germination of the heat-injured spores, (ii) prior combination with a component in a complex medium to prevent germination of heat-injured spores, (i...

Involvement of the spore coat in germination of Bacillus cereus T spores.

Bacillus cereus T spores were prepared on fortified nutrient agar, and the spore coat and outer membrane were extracted by 0.5% sodium dodecyl sulfate-100 mM dithiothreitol in 0.1 M sodium chloride (SDS-DTT) at pH 10.5 (coat-defective spores). Coat-defective spores in L-alanine plus adenosine germinated slowly and to a lesser extent than spores not treated with SDS-DTT, as determined by decreas...
